Let's break it down.

So this deck draws cards.

Spider-man draws a card every time he is attacked. Thus Taunt draws 4 cards. Coupled with Unflappable, that's 5. And if you can use Defensive Energy to play a defense event ("I Can Do This All Day", Desperate Defense, Never Back Down), that's a 6th card.

Nick Fury is also another way to draw cards.

So between Iron Fist, Mockingbird and Never Back Down and Webbed Up, you should be able to shut down the Villain.

At the speed you'll be drawing your deck, you'll be drawing additional Encounter cards. So keep Black Widow and Enhanced Spider-Sense on hand to mitigate the worst effects.

Peter's alter-ego generates 1 resources. This should help put out any of the following (in priority order); Ingenuity, Defensive Training, Unflappable, Aunt May, Defensive Conditioning, Ready for a Fight or Web-Shooter.

Ingenuity is a free resource which is just so universally useful during the life of a game

Defensive Training recycles your Protection cards.

Aunt May is probably one of the best cards in the game and if you can change form to Alter-Ego, gain advantage of your 6 card hand size and still defend against a Villain Attack by using Ready for a Fight (which will also net you an additional card!), this is a great start to your next turn.

In terms of card draw, Spider-man's inherent ability Spider Sense draws you a card every time you are attacked by the Villain.

Unflappable is an additional card and Defensive Energy also will allow you draw a card from playing any of "I Can Do This All Day", Desperate Defense or Never Back Down.

So what are the tactics with this deck aside from defending and drawing cards?

Simple: you want to shut down the Villain as much as possible.

Webbed Up therefore is your other go-to card. Did you know that you can either play Webbed Up on a stunned Villain or stun a Webbed Up Villain and that will shut them down for 3 attacks (Stun 1 -> prevents attack 1, then Webbed Up -> prevents attack 2, which drops a Stun -> preventing attack 3).

This one card can do so much heavy lifting.

In terms of Allies, Black Cat is part of your hero kit but she's pretty good. She doesn't incur consequential damage to Attack, so she is a free 1 damage for the rest of the game.

Black Widow is your emergency "get out of jail" to cancel a devastating encounter card. Between her and Enhanced Spider-Sense, you should be able to mitigate the game ending events.

Iron Fist lays down stuns. Stun + Webbed Up = Goodness.

And if Spidey does get in some trouble, you can always go home, get Aunt May to make you some chicken soup and then use Ready for a Fight to get back into the action.
