Spider-Man's biggest weaknesses in solo are his limited thwarting ability and his high capacity for dead hands (i.e., hands of cheap cards that you can't actually play). This deck tries to remedy that by keeping Spider-Man in hero form for as long as possible (so the villain never schemes), and filling the deck with cards you can always use on your turn.
You'll want to keep Black Widow in play indefinitely for her thwart, as well as her Interrupt ability, so use Med Team and First Aid to wipe her consequential damage, or to keep Spider-Man healthy enough to stay in the fight. Cards like Haymaker and Indomitable are good for turns when the rest of your hand is unusable (filled with cards like Counter-Punch or Enhanced Spider-Sense).
This deck is pretty flexible, and what you do with it does depend highly on which villain you're facing. Against Rhino, you mostly just want to pile on damage, using allies to keep threat low and soak up attacks; against Klaw, you want to build your engine up and keep control of the board with Enhanced Spider-Sense and Black Widow; and against Ultron, you want to keep him stunned/Webbed Up as much, so you can keep his drones away. I've been able to win fairly consistently against all three villains on Expert with this deck.
